There are many spiders which hunts about water, some can walk on water.
Just tweak this and that, put on a Proton Cannon and be done with it.

I voted Nature, b/c when I think of Spiders in fantasy I am always drawn back to this Magic the Gathering Card:

It was in my hand the first game I played (back around 1994), and though it lost to a Sengir Vampire, it helped defend my town. The concept of a giant spider being able to defend against flying units is so simple and natural, but fun and effective. I could see them in the nature town spinning webs to slow down other units, ground flying units (similar to a treant), and add complexity to the battle map so that ranged units can be better defended and used.

Neutral or nowhere, I say. Giant versions of real animals are always the least interesting units in factions like this (wolves, dogs, white tigers, bears, spiders, deer). No need to take away the lineup spot of a genuinely interesting mythical creature for yet another boring giant spider.

To those who say they would prefer a mythological creature over a giant animal, what if the animal fits the faction and lineup better than the mythological creature?

Except that apart from spiders, all of those animals you mention are at normal size, not enlarged.

They don't always exist, and sometimes when they do their names are so unpronounceable that they really have to be changes. Meso-American and Nordic seem pretty bad for that. There's also prenty of mythical creatures that are effectively just normal ones with special powers, like the Thunderbird.

Personally I love the idea of spiders in dungeon, or maybe even putting them in a Dark Elf town separated from dungeon.

Well, in my "ideal" version of Heroes, spiders belong to Coven faction, that is basically an alliance of swamp witches and all sorts of evil swamp and forest creatures. This faction combines evil, black magic with elements of wild nature, and I felt like spiders fit ideally there.
If we talk about more traditional factions, then I think Dungeon is the best fit. Narrow underground caverns fit for their habitat, plus there all sorts of underground creatures (like troglodytes) for spiders to eat. Plus, in "The Lord of the Rings" there was that giant spider living in the cave and since LotR is basically the staple of modern fantasy, it's not a shame to take some elements from it.
